Specificity and Comments:

The p53 homologue p63 encodes for different isotypes able to either transactivate p53 reporter genes (TAp63) or act as p53-dominant-negatives. The p63 is detected in prostate basal cells in normal prostate, however, is negative in malignant tumors of the prostate gland. Thus p63 is useful as a differential marker for benign and malignant tumors of prostate gland.

The high molecular weight (DE-SQ) recognizes cytokeratins 13, 14, 15, and 16. In normal prostate epithelia, DE-SQ positivity is confined to stratified epithelia, myoepithelial cells and basal cells in the prostate gland and bronchi. It stains virtually identical to 34Eß12. The combination of HMW CK and p63 has been shown to be superior that each alone. The basal cell cocktail not only increases the sensitivity of the basal cell detection, but also reduces the staining variability and therefore renders the basal cell immunostaining more consistent (see important reference on backside).
